Holstein steers (n = 40; initial BW = 84.9 ±â€…7.1 kg) were used to study the genesis of liver abscesses (LA) using an acidotic diet challenge with or without intraruminal bacterial inoculation. Steers were housed in individual pens inside a barn and randomly assigned to one of three treatments: (1) low-starch control diet comprised primarily of dry-rolled corn and wet corn gluten feed (CON); (2) high-starch acidotic diet with steam-flaked corn (AD); or (3) acidotic diet plus intraruminal inoculation with Fusobacterium necrophorum subsp. necrophorum (9.8 × 108 colony forming units [CFU]/mL), Trueperella pyogenes (3.91 × 109 CFU/mL), and Salmonella enterica serovar Lubbock (3.07 × 108 CFU/mL), previously isolated from LA (ADB). Steers in AD and ADB were fed the acidotic diet for 3 d followed by 2 d of the CON diet, and this cycle was repeated four times. On day 23, ADB steers were intraruminally inoculated with the bacteria. At necropsy, gross pathology of livers, lungs, rumens, and colons was noted. Continuous data were analyzed via mixed models as repeated measures over time with individual steer as the experimental unit. Mixed models were also used to determine the difference in prevalence of necropsy scores among treatments. Ruminal pH decreased in AD and ADB steers during each acidotic diet cycle (P ≤ 0.05). LA prevalence was 42.9% (6 of 14) in ADB vs. 0% in AD or CON treatments (P < 0.01). Ruminal damage was 51.1% greater in ADB than in AD (P ≤ 0.04). Culture of LA determined that 100% of the abscesses contained F. necrophorum subsp. necrophorum, 0% contained T. pyogenes, 50% contained Salmonella, and 50% contained a combination of F. necrophorum subsp. necrophorum and Salmonella. The F. necrophorum subsp. necrophorum was clonally identical to the strain used for the bacterial inoculation based on phylogenetic analysis of the whole genome. This experimental model successfully induced rumenitis and LA in Holstein steers and confirms the central dogma of LA pathogenesis that acidosis and rumenitis lead to the entry of F. necrophorum into the liver to cause abscesses. Our findings suggest that an acidotic diet, in conjunction with intraruminal bacterial inoculation, is a viable model to induce LA. Further research is needed to determine the repeatability of this model, and a major application of the model will be in evaluations of novel interventions to prevent LA.


Liver abscesses (LA) in feedlots are costly to the beef industry. At harvest, LA cause an increase in liver condemnations, carcass trimming, and a decrease in quality grade. The objective of this research was to develop an experimental LA model in Holstein steers using an acidotic diet with and without intraruminal inoculation of bacteria involved in LA formation. These data suggest acidotic diet challenges in conjunction with bacterial inoculation were able to induce LA in Holstein steers. The acidotic diet alone caused reduced rumen content pH and caused rumen wall inflammation and damage, observed at harvest. Nonetheless, the addition of bacteria had a compounding effect on rumen damage. Both bacteria inoculated were isolated from 57% of LA suggesting they may work in synergy to form LA.

The effects of a novel direct-fed microbial (DFM) on feedlot performance, carcass characteristics, digestibility, ruminal morphology, and volatile fatty acid (VFA) profile of finishing steers were evaluated. Single-source Angus-crossbred yearling steers (n = 144; initial body weight (BW) = 371 ±â€…19 kg) were used in a randomized complete block design. Steers were blocked by initial BW and randomly assigned to treatments (12 pens/treatment; 4 steers/pen). Treatments included (A) CONTROL (no DFM, tylosin, or monensin, (B) MONTY (monensin sodium [330 mg/animal-daily] and tylosin phosphate [90 mg/animal-daily]), and (C) MONPRO (monensin sodium [same as previous] and Lactobacillus salivarius L28 [1 × 106 CFU/animal-daily]). Treatments were included in a steam-flaked corn-based finisher diet offered once daily using a clean-bunk management for ~149 d. The digestibility assessment was performed from days 70 to 74. Ruminal fluid and rumen tissue samples were collected at the slaughter for VFA profile and papillae morphology analyses, respectively. Data were analyzed using the GLIMMIX procedure of SAS with pen serving as the experimental unit, treatment as fixed effect, and BW block as random effect. Steers offered MONPRO had on average 5.3% less (P < 0.01) dry matter intake (9.56 kg/d) compared with either CONTROL (10.16 kg/d) or MONTY (9.96 kg/d). The carcass-adjusted final BW (613 kg; P = 0.23), overall average daily gain (1.64 kg/d; P = 0.23), and gain-efficiency (0.165; P = 0.61) were not affected by treatments. Steers offered CONTROL had greater (P < 0.01) marbling score and tended (P = 0.06) to have less carcasses grading Select and tended (P = 0.10) to have more carcasses grading Upper-Choice, while other carcass characteristics and liver-abscesses were not affected (P ≥ 0.23) by treatments. The digestibility of nutrients (P ≥ 0.13) and the ruminal VFA profile (P ≥ 0.12) were not affected by treatments. Steers offered MONPRO tended (P = 0.09) to have 16% greater average papillae number compared to other treatments. Yearlings offered finishing diets containing L. salivarius L28 plus monensin did not affect growth performance, digestibility, or ruminal VFA, but reduced feed intake. Carcass quality was negatively affected by treatments, while animals consuming L. salivarius L28 and monensin tended to improve ruminal morphology. Current findings in ruminal morphology and feed intake may warrant further assessment of diets containing L. salivarius L28 on beef cattle food safety aspects.


Antimicrobial resistance is a growing concern to public health and medically important antibiotics have been listed in the Veterinary Feed Directive. Nutritional technologies, such as direct-fed microbials, are being increasingly studied for the development of an effective use on beef cattle production systems. The newly isolated strain of Lactobacillus salivarius L28 has demonstrated pathogenic inhibition of Escherichia coli, Salmonella, and Listeria monocytogenes on in vitro assessments. The potential benefits have warranted the exploration of L. salivarius L28 in a feedlot setting. Single-source Angus-crossbred yearling steers were offered steam-flaked corn-based finishing diets containing no feed additive, or either a combination of tylosin plus monensin or L. salivarius L28 plus monensin. Steers offered L. salivarius L28 plus monensin consumed 5.3% less feed compared with other treatments, while other growth performance variables and the digestibility of nutrients were not affected. Carcasses from cattle supplemented with monensin had slightly lower carcass quality grades than those not supplemented with monensin. Lactobacillus salivarius L28 plus monensin tended to improve steers ruminal morphology. Current findings may warrant further food safety assessments when cattle are offered diets containing L. salivarius L28.

Holstein steers (n = 40; initial body weight [BW] = 96.0 ± 10.5 kg) were individually housed in a climate-controlled barn to evaluate potential models for the genesis of liver abscesses (LA). In this 2 × 2 factorial, steers were balanced by BW and randomly assigned to one of two treatments: 1) intravenous saline injection followed by intraruminal bacterial inoculation with Fusobacterium necrophorum subsp. necrophorum (1 × 109 colony forming unit [CFU]/mL) and Salmonella enterica serovar Lubbock (1 × 106 CFU/mL; CON; n = 20 steers); or 2) intravenous injection with 0.25 µg/kg BW of lipopolysaccharide (LPS; Escherichia coli O111:B4) followed by intraruminal bacterial inoculation of F. necrophorum subsp. necrophorum (1 × 109 CFU/mL) and S. enterica serovar Lubbock (1 × 106 CFU/mL; LBI; n = 20 steers) and 1 of 2 harvest dates (3 or 10 d post LPS infusion). Body weights were recorded on days -4, -1, 3, and 10, and blood was collected for hematology on days -4, 3, and 10, relative to LPS infusion on day 0. Intraruminal bacterial inoculation occurred on day 1. Steers from each treatment group were harvested at two different time points on day 3 or 10 to perform gross pathological examination of the lung, rumen, liver, LA (if present), and colon. Feed disappearance was less for LBI than CON (P < 0.01); however, BW did not differ (P = 0.33) between treatments. Neither treatment nor time differed for hematology (P ≥ 0.13), and no gross pathological differences were noted in the lung, liver, LA, or colon (P ≥ 0.25). A treatment × harvest date interaction was noted for ruminal pathology in which LBI had an increased percentage of abnormal rumen scores on day 3 (P < 0.01). These results suggest that an LPS challenge in combination with intraruminal bacterial inoculation of pathogens commonly isolated from LA was not sufficient to induce LA in steers within 3 or 10 d (P = 0.95) when compared to CON. Further evaluation is needed to produce a viable model to investigate the genesis and prevention of LA in cattle.


Liver abscesses in feedlot cattle can cause a decrease in feed intake, average daily gain, feed efficiency, and hot carcass weight. At harvest, liver abscesses result in liver condemnations, carcass trimming, and a potential decrease in quality grade, with an estimated economic cost to packers of $41.6 million annually. Our objective was to evaluate an intravenous endotoxin challenge followed by intraruminal inoculation of bacteria commonly isolated from liver abscesses over a 10-d period as a potential model to understand the genesis and etiology of liver abscesses in cattle and evaluate possible preventative interventions. Results suggest that an endotoxin challenge in combination with intraruminal bacterial inoculation is not a viable model to induce liver abscesses in steers, and bacterial inoculation alone was insufficient to induce liver abscesses. The length of time necessary to induce liver abscesses is also unknown. Based on our results, more research is needed to develop a noninvasive model to induce liver abscesses in cattle.

Heat stress abatement strategies for pre-weaned dairy calves are seldom evaluated. An experiment was conducted to evaluate the effects of housing calves under a barn and provision of fans to calves housed under a barn on calfhood performance. The experiment was conducted in a dairy in southern Georgia, USA. Male Holstein calves (n = 60; 0 to 68 day of age) were assigned randomly at birth (day 0) to 1 of 3 treatments: hutch outdoors with 50% of its area covered with plywood (control = 20), hutch in a barn with no cooling (SH = 21), and hutch in a barn with ceiling fans (SHF = 19). Body weight (BW) was measured at birth, and total serum protein and wither-height were measured 24 to 48 h after birth. A sub-set of hutches was evaluated for air speed and temperature, and rectal temperature (RT) and respiratory frequency (RF) of calves housed in these hutches were measured at 0900 and 1500 h. Intakes of liquid feed (days 14 to 63) and starter (days 14 to 68) were recorded daily, BW and wither-height were measured weekly, and feed efficiency was calculated weekly. Blood was sampled on days 1, 14, 28, 42, 49, 52, 56, 58, 63, and 65 for the measurement of fatty acids, ß-hydroxybutyrate, glucose, and insulin. The SHF treatment resulted in air velocity 0.56 to 0.83 m/s greater (P < 0.01) than the control and SH treatments, respectively, whereas the control treatment resulted in air temperature 1.2 to 3.2 °C greater (P < 0.01) than the SH and SHF treatments, respectively. The RT of calves in the control treatment was 0.1 to 1.1 °C greater (P ≤ 0.03) than the SH and SHF treatments, respectively, and the control treatment resulted in RF 39.4 to 60.2 mov/min greater (P < 0.01) than the SH and SHF treatments, respectively. Treatment did not (P ≥ 0.27) affect feed efficiency and concentrations of metabolites and insulin, but calves in the control treatment were 2.6 cm shorter (P = 0.03) than calves in the SHF treatments at weaning. Provision of fans to calves housed under a barn reduced RT, RF, but only had a minute impact on wither-height.

The objective of this study was to determine the effects of supplementing a commercial porous ceramic clay particle, with or without a blend of preservatives, on the performance and nutrient digestibility of weanling pigs. Fifteen weanling pigs of the Yorkshire, Landrace, and Duroc breeds were blocked by breed and randomly assigned to one of three treatments (n = 5): (1) Control, non-medicated diet with no additional feed additives (CON); (2) PowerGuard, basal diet with 0.25% of the DM consisting of a ceramic particle mixed into the pelleted feed (PG; MB Nutritional Sciences, Lubbock, TX, 79403); or (3) Power Guard + a blend of preservatives, basal diet with 0.3% of the DM consisting of the ceramic clay and preservatives mixed into the pelleted feed (PG-D). The facility was temperature controlled with an average temperature of 28.5 °C. Pigs were offered ad libitum access to feed and water and were housed individually in elevated crates. Body weights were collected upon enrollment on day 0 and at the end of the observation period on day 18. On day 15 , a 72-h total feed and fecal collection period began. Feed and fecal samples were analyzed for DM, CP, Ash, OM, ADF, NDF, zinc, copper, thiamin (vitamin B1), and retinol (vitamin A). Liver samples were collected immediately after harvest and frozen for later mineral analysis. Data were analyzed using Proc Mixed in SAS with dietary group as the main effect and block as the random effect (SAS 9.4, Cary, NC). There were no treatment differences in performance measures including final BW, ADG, or G:F (P ≥ 0.701). There were no treatment differences in diet nutrient digestibility for DM, CP, Ash, OM, ADF, or NDF (P ≥ 0.312). Additionally, there were no treatment effects on zinc, copper, or retinol digestibility (P ≥ .298); however, thiamin inclusion rate was increased for the PG-D treatment, thus leading to an increased digestibility for thiamin (P = 0.018) in the PG-D treatment. There were no treatment differences in hepatic mineral concentrations (P ≥ 0.532); however, there was a tendency for pigs fed PG-D to have increased hepatic concentrations of lead and mercury when compared with both PG and CON pigs (P ≤ 0.066). In summary, supplementation of a commercial ceramic particle with or without a blend of preservatives to weaned pigs did not affect performance or apparent nutrient digestibility.

Genetic and reproductive advancements in the dairy industry, volatile milk markets, and beef packer restrictions on dairy carcasses have increased the popularity of crossbreeding beef sires to dairy cows in the United States. This observational study aimed to understand performance of dairy cows bred to beef sires and feedlot and carcass performance of crossbred beef × dairy cattle. For dairy cow performance, archived records from two dairies representing two successive lactations were evaluated in cows (Dairy A: n = 72/group; Dairy B: n = 456/group) representing 1) All Dairy, where previous sire type of conception was Holstein for both lactations, or 2) Beef on Dairy, where previous sire type of conception was Holstein for the preceding lactation and a beef breed for the subsequent lactation. For feedlot performance, closeout data from pens (n = 26/cattle type) of beef and beef × dairy steers and heifers were evaluated. For carcass performance, individual carcass data were compared between conventional beef (n = 966), beef × dairy (n = 518), and Holstein (n = 935) steers sampled across a variety of processing facilities, harvest lots, and geographical regions. Cow lactation performance was minimally impacted by sire type of previous conception. Cows conceived to beef sires exhibited a 2 to 3 d greater (P < 0.01) gestation length than cows conceived to Holstein sires. Beef × dairy cattle were not largely different in weight gain at the feedlot but exhibited 1-unit lesser (P < 0.01) dressing percentage than beef cattle. Beef × dairy carcasses possessed 18% lesser (P < 0.05) 12th rib fat thickness than beef cattle and 5% greater (P< 0.05) ribeye area than dairy cattle. Additionally, beef cattle produced nearly double (P < 0.05) the percentage of yield grade 4 carcasses produced by beef × dairy and Holstein cattle.

We determined effects of pre- and postpartum dietary cation-anion difference (DCAD) on urine pH, serum calcium and hormone concentrations, and milk production with 48 multiparous Holstein cows (average body weight = 706 ± 7.3 kg). Treatments were 3 prepartum DCAD concentrations (0, -100, or -180 mEq/kg dry matter [DM]) and 2 postpartum DCAD concentrations (+250 or +350 mEq/kg DM) starting 29 days before parturition through 90 days in milk. Prepartum urine pH was lower (p < 0.05) for -180 than for -100 or 0 DCAD, and postpartum urine pH was higher (p < 0.05) for +350 than for +250 DCAD. Prepartum serum total and ionized calcium and hydroxyproline were greater (p < 0.05) for -180 than for -100 and 0 DCAD, whereas parathyroid hormone was greater (p < 0.05) for 0 than for -100 and -180 DCAD. After calving, negative prepartum DCAD increased (p < 0.05) serum total and ionized calcium, but effects varied by sampling day. Pre- and postpartum DCAD did not affect milk yield or milk fat, but milk protein percent and total solids were increased (p < 0.05) by negative prepartum DCAD. Feeding an acidogenic diet prepartum improved postpartum calcium status without major effects on milk yield and composition.

In the feedlot, there can be a decrease in dry matter intake (DMI) associated with reimplanting cattle that negatively affects growth performance. This study was conducted to determine the mechanisms causing a decrease in DMI after reimplanting and identify a strategy to mitigate the decrease. Crossbred steers (n = 200; 10 pens/treatment; initial bodyweight [BW] = 386 ±â€…4.9 kg) were used in a randomized complete block design experiment. Cattle were implanted with Revalor-IS on day 0. Treatments included a Revalor-200 implant on day 90 before feeding with the following management practices imposed: 1) steers were returned to their home pen immediately after reimplant (PCON); 2) steers were placed in pens and restricted from feed and water for 4 h (RES); 3) steers were walked an additional 805 m after reimplant and then returned home (LOC); 4) steers were restricted from feed and water for 4 h and walked an additional 805 m (RES + LOC); 5) steers were given an oral bolus of Megasphaera elsdenii (Lactipro; MS Biotec, Wamego, KS) and were restricted from feed and water for 4 h, and then walked an additional 805 m (LACT). One hundred steers were given an ear tag to record minutes of activity (ESense Flex Tags, Allflex Livestock Intelligence, Madison, WI). As a percentage of BW, DMI was 5% greater (P = 0.01) from reimplant to end for PCON vs. RES, LOC, and RES + LOC treatments. Likewise, as a percentage of BW, DMI was 6.6% greater (P = 0.03) from reimplant to end and 4.0% greater (P = 0.05) overall for the PCON treatment vs. the LOC treatment. Overall, DMI as a percentage of BW was 3.3% greater (P = 0.02) for PCON vs. RES, LOC, and RES + LOC treatments. There was an increase in G:F from reimplant to end (P = 0.05) for RES + LOC vs. the LACT treatment. From these data, we conclude that restricting cattle from feed and water for 4 h after reimplanting did not alter subsequent DMI. Increasing locomotion had the greatest negative effect on DMI and growth performance. Management strategies to decrease locomotion associated with reimplanting would be beneficial to DMI and overall growth performance of finishing beef steers.

In this literature review, we overview some of the common management practices associated with calf rearing in specialized operations of the United States. Given the growing importance of dairy-beef calves entering the beef production of the United States, we overview aspects related to housing, nutrition, and health events during the pre- and post-weaning period. Based on data on dairy animals, we hypothesize how early life experiences could impact the feedlot performances of dairy-beef animals. Most of the large calf raising operations, where the majority of dairy-beef animals are raised, are located in the Central Great Plains and West regions of the United States. Approximately 80% of calves are individually housed, but the type of housing (e.g., outside hutch, inside a barn) varies based on location of calf-raising facilities. Milk-replacer is fed in more than 80% of operations, while milk (saleable or nonsaleable) is fed in approximately 30% of calf raising facilities (some operations fed more than one type of liquid diet). In addition to liquid feed, water and calf starter are offered ad libitum to calves. Adequate starter intake at weaning is crucial for feed transition from pre- to post-weaning period, which occurs at approximately 2 months of age. Then, calves are mainly housed in group pens and transition from calf-starter to total mixed ration (TMR). Health challenges such as scours and bovine respiratory disease (BRD) can hinder the performance of calves and are major causes of morbidity and mortality in calf ranches. Transportation at a very young age and comingling with animals from other dairies can increase the risk of diseases. Current research efforts are focusing on determining individual factors such as body weight (BW) at arrival or biomarkers of inflammation and stress that can be predictive of disease morbidity, mortality, and performance of calves. Future research should focus on how to utilize this information to optimize management and to develop targeted preventative strategies to reduce incidence of diseases and mortality and improve performance during the pre-weaned period. Also, more research is needed to understand how colostrum management, housing, and nutrition can impact the adult performance of dairy-beef animals.

This experiment evaluated forage quality, total nutrient yield, water footprint, and growth performance of beef steers receiving protein supplements while grazing Teff grass ['Tiffany'Eragrostis tef (Zucc.) Trotter] over two consecutive growing seasons. Each year, four 2.66-ha irrigated paddocks (experimental units) were stocked with crossbred beef steers (n = 5 per paddock, initial BW = 289 ± 30 for yr 1; and n = 6, initial BW = 286 ± 23 for yr 2) in a randomized complete block design and stocked continuously for 63 d. Daily supplements [0.45 kg/d of cottonseed meal (Control) enough to avoid a negative ruminal N balance; and 0.50% mean paddock BW animal-daily (approximately 1.65 kg) of sorghum-dried distillers grains plus solubles, (DDGS)] were randomly assigned to two paddocks each. Supplement did not influence forage neutral detergent fiber (NDF), acid detergent fiber, crude protein, or in vitro true digestibility (P ≥ 0.54), except for a tendency (P = 0.08) for a numerical increase in NDF content of paddocks with steers that received DDGS supplementation. Paddock nutrient-yields were similar (P ≥ 0.43) between supplement treatments. Supplementation with DDGS produced greater (P = 0.01) cattle shrunk average daily gain (ADG). Predicted teff dry matter intake (DMI), net energy for maintenance (NEm), and growth (NEg) (P ≤ 0.03) were greater with cattle offered Control treatment. Predicted total DMI was similar (P = 0.14) although predicted dietary NEm, NEg, gain:feed, and total BW gain were greater (P ≤ 0.02) with DDGS. Predicted forage intake was greater (P ≤ 0.05) for cattle offered Control treatment. Teff nutrients remaining on d 56 were similar (P = 0.33) between treatments. Water footprint for total production of forage nutrient components did not differ (P ≥ 0.12) by treatments. Nutrient yield and water use efficiency of continuously stocked teff grass was not affected by supplemental regimen. Using DDGS as a supplement may increase BW gain through increased nutrient utilization without hindering teff nutrient production on a continuous stocking system.

Effects of dried distillers grains plus solubles (DDGS) on ruminal fermentation, degradation kinetics, and feeding behavior of steers offered annual (Eragrostis tef; TEFF) or perennial (Bothriochloa bladhii; OWB) grass hay were evaluated. Ruminally cannulated Angus crossbred steers (n = 6; body weight [BW] = 304 ± 11 kg) were assigned to a 4 × 6 unbalanced Latin square design with four treatments arranged as a 2 × 2 factorial: hay type (OWB or TEFF) and DDGS supplementation (0% or 0.5% BW [dry matter {DM} basis]). Steers had ad libitum access to hay. Periods consisted of a 14-d adaptation followed by 7 d of collection. Residues from the in situ incubations (0, 3, 6, 12, 24, 36, 48, 72, and 96 h post-feeding) were fitted to a first-order kinetics model using the NLIN procedure of SAS. The DDGS decreased (P < 0.01) TEFF DM intake (DMI) by 11.3%, while not affecting DMI of OWB. The greatest DMI was observed for steers supplemented with DDGS, regardless of forage, and least in steers consuming OWB without DDGS (hay type × DDGS; P = 0.03). Non-supplemented steers spent more (P < 0.01) time eating hay. Digestibility of DM tended (P = 0.06) to increase with DDGS supplementation. A hay type × DDGS interaction was observed (P ≤ 0.05) on ruminal effective degradable fractions. The rate of degradation, soluble fraction, and the potentially degradable fraction of organic matter (OM), neutral detergent fiber, and acid detergent fiber (ADF) increased (P ≤ 0.05), while the undegradable fraction of all components decreased (P ≤ 0.01) when steers were offered TEFF compared to OWB. Ruminal DM, OM, and ADF degradation lag-time increased (P ≤ 0.02) in steers offered OWB. Ruminal degradation kinetics were not (P ≥ 0.17) independently affected by DDGS supplementation. Average ruminal pH of steers offered TEFF (P < 0.01) and those offered DDGS (P < 0.01) were lower than OWB and non-supplemented steers. Total concentration of VFA tended (P = 0.09) to increase when DDGS was provided with OWB, while decreasing when TEFF was offered. The acetate:propionate increased (P < 0.01) with DDGS supplementation due to a decrease (P = 0.03) in propionate. Ruminal NH3-N was greater (P = 0.03) in steers offered TEFF compared to OWB, and those supplemented with DDGS (P = 0.03). An annual, in place of a conventional, perennial hay improved intake and digestion of nutrients, without affecting feeding behavior. The supplementation with DDGS appears to affect forage intake, ruminal degradation, and feeding behavior, although not independent of forage quality.

The objective of this study was to evaluate the efficacy of two metaphylactic strategies using tildipirosin for the control of bovine respiratory disease (BRD) in dairy calves transported to a heifer raising facility within their first week of life. A total of 2,100 calves were enrolled in the study. Animals were transported for ~1,715 km, from dairies located in Minnesota to a calf raising facility located in New Mexico, where they were housed in individual hutches until weaning. Three days after arrival, calves were randomly allocated into three groups: (1) META1: single subcutaneous (SQ) injection of tildipirosin (Zuprevo™, Merck Animal Health) at enrollment at 4 mg/kg; (2) META2: SQ injection of tildipirosin at enrollment and 17 days later; (3) CON: untreated controls. The BRD incidence was 11.4, 10.8, and 9.4% for calves enrolled in the CON, META1, and META2, respectively (P = 0.44). Lung lesions diagnosed through ultrasonography was found in 21.0, 21.0, and 21.8% of calves enrolled in CON, META1, and META2, respectively (P = 0.99). Mortality tended to be greater for CON calves in comparison to META2 calves (1.5 vs. 0.6%, P = 0.06), but did not differ between calves enrolled in CON and META1 groups (1.5 vs. 1.2%, P = 0.55). Growth was not affected by metaphylaxis. The average daily gain for calves enrolled in CON, META1, and META2 was 517, 518 and 525 g, respectively (P = 0.25). Blood analysis revealed that some of the markers of inflammation assessed were influenced by metaphylaxis. At 27 days after enrollment, META2 calves had decreased concentrations of haptoglobin, serum amyloid A, and aspartate aminotransferase, compared to CON calves (P < 0.05). Additionally, CON calves had increased concentrations of globulins and lower albumin to globulin ratio than META2 calves at the end of the weaning period (P < 0.05). In conclusion, tildipirosin metaphylaxis did not decrease the incidence of BRD nor did it have an impact on weight gain. However, metaphylaxis with two injections of tildipirosin at enrollment and 17 days later tended to reduce mortality and improved the systemic inflammatory status of calves during the preweaning period.

Available energy plays a critical role in the initiation and maintenance of an immune response to a pathogen, a process that is further altered by activation of the stress system. This study was designed to determine the effect of an acute vs chronic stress model on the metabolic response to vaccination in naïve beef steers. Steers (n = 32; 209 ± 8 kg) were blocked by body weight (BW) and randomly assigned to one of three treatments: 1) Chronic stress (CHR), 0.5 mg/kg BW dexamethasone (DEX) administered i.v. at 1000 h on day 3 to day 0; 2) Acute stress (ACU), 0.5 mg/kg BW DEX administered i.v. at 1000 h on day 0 only; or 3) Control (CON), no DEX. On day -4, steers were fitted with jugular vein catheters and moved into individual bleeding stalls in an environmentally-controlled facility. Blood samples were collected at -74, -50, and -26 h, at 0.5-h intervals from -4 to 6 h, and at 12, 24, 36, 48, and 72 h relative to vaccination with a combination vaccine (Pyramid 5 + Presponse SQ, Boehringer Ingelheim Animal Health USA, Duluth, GA) at 1200 h on day 0. Data were analyzed by the MIXED procedure of SAS specific for repeated measures. There was a treatment × time interaction (P < 0.001) for serum glucose concentrations. Specifically, glucose concentrations increased at -50 h in CHR steers and at 1200 h in ACU steers and remained elevated through 72 h postvaccination period in these two treatments compared to CON steers. The change in nonesterified fatty acid (NEFA) concentrations relative to baseline values was affected by treatment and time (P < 0.001) such that the change in NEFA was greater in CHR (0.06 ± 0.01 mmol/L), followed by CON (-0.01 ± 0.01 mmol/L) and ACU steers (-0.04 ± 0.01 mmol/L). There was a tendency (P = 0.08) for a treatment × time interaction for change in serum NEFA concentrations. Serum urea nitrogen (SUN) was affected by treatment and time (P < 0.001) such that SUN concentrations were greatest in CHR (12.0 ± 0.1 mg/dL) followed by ACU (10.4 ± 0.1 mg/dL) and CON steers (9.6 ± 0.1 mg/dL); however, the treatment × time interaction was not significant (P = 0.12). These data demonstrate that activation of the stress and immune axes using an acute or chronic stress model can increase energy mobilization prior to and following vaccination in naïve steers, potentially affecting available energy needed to mount an adequate antibody response to vaccination.

The study objective was to determine if a combined weaning and transportation stress model affected performance, antibody, endocrine, or hematological responses to modified-live virus (MLV) or killed virus (KV) respiratory vaccination in beef steers. In total, 48 calves (Day 0 BW = 226 ± 6.2 kg) from a single origin were used in a 2 × 2 factorial to evaluate main effects of stress model, vaccine type, and their interaction, resulting in four treatments (n = 12/treatment) including non-stress control (C) with KV (CKV), C with MLV (CMLV), stress model implementation (S) with KV (SKV), and S with MLV (SMLV). The C calves were weaned at the origin ranch on Day -37 and transported 472 km to the study site on Day -21 to allow acclimation. The S calves were weaned on Day -3, transported 460 km to a research facility on Day -2, held overnight, and transported 164 km to the study site on Day -1 to mimic the beef cattle marketing process. Vaccines were administered on Day 0 and KV was revaccinated on Day 14. The animal was the experimental unit and dependent variables were analyzed using PROC MIXED with repeated measures (day). A stress model effect (p = 0.01) existed for DMI from Day 0 to Day 7 with greater DMI for C (6.19 vs. 4.64 kg/day) when compared to S. The MLV groups had reduced (p = 0.05) ADG from Day 0 to Day 56, compared to KV. There was a vaccine type × day (p < 0.01) interaction with increased (p ≤ 0.01) PI3V- and IBRV-specific antibody titers for KV on Day 21; conversely, MLV had increased (p ≤ 0.01) BVDV titers on Days 14, 28, 35, 42, 49, and 56. Increased (p ≤ 0.05) BRSV titers were observed in a stress model × day (p < 0.01) interaction for S on Days 21, 28, 36, and 42; however, C exceeded S in BVDV-specific antibody concentration on Days 21, 28, and 49. A day effect (p < 0.01) was observed for serum haptoglobin with the greatest (p < 0.01) concentration on Day 3. Serum cortisol concentration was greater (p ≤ 0.04) for C vs. S on Days -2, 0, 1, 3, and 5. Total leukocytes were decreased for C vs. S on Days 0, 1, 3, 5, 7, 14, and 21 (p ≤ 0.02). A reduction (p ≤ 0.04) in total leukocytes was observed for MLV on Days 5, 7, and 14 vs. KV. Neutrophils and neutrophil:lymphocyte were markedly increased (p ≤ 0.01) for S on Day -2, whereas neutrophils were decreased (p ≤ 0.01) on Days 1 and 21 for S. Monocytes were decreased on Days 1, 5 and 7 for MLV (p ≤ 0.04) and Days -2 to 14 for S (p ≤ 0.03). Eosinophils were reduced (p = 0.007) for S vs. C on Day -2, yet a distinct rebound response (p = 0.03) was noted for S on Day 0. The results indicate that S and MLV vaccination more profoundly induced immunomodulation in beef calves.

OBJECTIVE: To quantify acute immunologic and metabolic responses of beef heifers following topical administration of transdermal flunixin meglumine (TDFM) at various times relative to bovine herpesvirus 1 (BHV1) and Mannheimia haemolytica challenges. ANIMALS: 32 beef heifers (mean body weight, 170 kg). PROCEDURES: Heifers were assigned to 1 of 4 groups. Heifers in the control group did not receive TDFM, whereas 1 dose of TDFM (3.3 mg/kg) was topically applied to heifers of groups A, V, and B at -144, -72, and 0 hours. All heifers were inoculated with 1 × 108 plaque-forming units of BHV1 in each nostril at -72 hours and with 1.18 × 106 CFUs of M haemolytica intratracheally at 0 hours. Vaginal temperature was recorded and blood samples were collected for quantification of select immunologic and metabolic biomarkers at predetermined times from -144 to 360 hours. RESULTS: Mean vaginal temperature was similar between group A and the control group. Mean vaginal temperatures for groups V and B were generally lower than that for the control group following BHV1 and M haemolytica challenges, respectively. Mean neutrophil oxidative burst capacity and L-selectin expression at 0 hours were significantly decreased for group V relative to the other groups. Other biomarkers did not differ among the groups at any time. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that topical administration of TDFM to beef cattle effectively alleviated pyrexia without adverse effects on acute immunologic or metabolic responses when TDFM was administered at the same time as, but not before, respiratory pathogen challenge.

The use of portable X-ray fluorescence (PXRF) spectrometry to detect external markers on processed or unprocessed cattle and sheep fecal specimens to estimate apparent total tract digestibility (ATTD) was evaluated. Exp. 1: ruminally cannulated Angus-crossbred steers (n = 7; BW = 520 ± 30 kg) were individually fed ad libitum for 21 d in a completely randomized design (CRD). Markers (Cr2O3 and TiO2) were placed inside the rumen twice daily (7.5 g of each marker). Fecal samples were collected twice daily from day 14 to 21. Exp. 2: crossbred wethers (n = 8; BW = 68 ± 3 kg) were individually fed ad libitum for 21 d in a CRD. During this period, 2 g of Cr2O3 and TiO2 were top-dressed onto the feed twice daily. Sheep were housed in metabolism crates for 5 d for total fecal collection. Concentration of markers was determined on diets, refusals, and fecal specimens (fresh, dry-only, and dried/ground) using atomic absorption to detect Cr and spectrophotometry for Ti. Concentration of both markers was also determined via the PXRF spectrometer. Delta between ATTD estimated by wet chemistry and PXRF was not different from zero (P ≥ 0.14) when using cattle fresh fecal specimens for both markers, whereas ATTD estimated by PXRF with dry-only and dried/ground fecal specimens were 3.6 and 1.1 percent units lower (P ≤ 0.04), respectively, than ATTD estimated by wet chemistry for Cr and Ti, respectively. Regardless of the fecal sample preparation method on cattle specimens, Ti concentration was similar (P = 0.39) among methodologies, while Cr was underestimated (P < 0.01) by 13% when PXRF was used in dry-only or dried/ground samples. The ATTD of sheep was underestimated (P < 0.01) by 2.4 percent units compared with control when Cr was measured by PXRF in dry-only samples. The Cr concentration in dry-only fecal specimens of sheep tended (P = 0.09) to be lower compared with wet chemistry analysis. Fresh and dry/ground sheep fecal samples assessed for Cr, and dry-only assessed for Ti were not (P ≥ 0.49) affected by detection method. The Cr fecal recovery tended (P = 0.10) to be the lowest for dry-only, the greatest for wet chemistry, intermediate for fresh and dry/ground sheep-fecal specimens; while not affected (P = 0.40) for Ti. The PXRF is an accurate technology to detect Cr and Ti in fresh cattle fecal samples to estimate ATTD. For fresh and dry/ground, the technology was effective for determining the concentration of Cr, or dry-only fecal specimens when detecting Ti in sheep specimens.

Livestock industries strive to improve the health of their animals and, in the future, they are going to be required to do this with a continued reduction in antimicrobial use. Nutraceuticals represent a group of compounds that may help fill that void because they exert some health benefits when supplemented to livestock. This review is focused on the mechanisms of action, specifically related to the immune responses and health of ruminants. The nutraceutical classes discussed include probiotics, prebiotics, phytonutrients (essential oils and spices), and polyunsaturated fatty acids.

The effects of dietary pretreatment with fibrolytic enzyme-based cocktail were evaluated in 2 studies: (1) in vitro true digestibility; and (2) intake, digestibility, feeding behavior, and ruminal fermentation of beef steers fed growing diets. For the in vitro assessment, the ruminal inoculum was collected from 2 steers (BW = 543 ± 45 kg; 4-h after feeding; growing diets) and enzymes included or not (Trichoderma reesei fermentation extract; 0.75 µL/g of substrate DM). Within in vitro batches (n = 4), 12 substrates were incubated and in vitro true nutrient digestibility was evaluated. For study 2, 5 ruminally cannulated beef steers (BW = 520 ± 30 kg) were used in a 5 × 4 unbalanced Latin square using a 2 × 2 factorial arrangement of treatments: (a) diet quality (high = HQ; and low = LQ) and (b) enzyme inclusion (0 or 0.75 mL/kg of diet DM). Steers were fed ad libitum during four 21-d periods consisting of 14-d of adaptation and 7-d of collections. An enzyme × substrate was observed (P < 0.01), in which DM, OM, and NDF disappearance of sorghum grain increased with enzymes addition. Addition of enzymes increased (P < 0.01) ADF digestibility for all substrates. No diet quality × enzyme (P ≥ 0.18) was observed for intake variables in study 2. Enzyme-fed steers increased (P ≤ 0.05) intake of DM, digestible DM, NDF, and ADF compared with steers not fed fibrolytic enzymes. Addition of enzyme did not affect (P ≥ 0.28) apparent total tract digestibility of beef steers. Steers fed HQ diets consumed more (P ≤ 0.04) DM, digestible DM and OM, and less (P ≤ 0.03) total and digestible fiber than steers fed LQ diets. Ruminal pH average decreased (P = 0.01) for steers fed HQ or enzyme-fed diets compared with other treatments. A tendency (P = 0.06) toward improved total VFA was observed on enzyme-fed steers with HQ diets, but not for LQ diets. The molar proportion of ruminal propionate increased (P = 0.01) when steers were fed enzyme. Steers fed HQ diets had greater (P < 0.01) propionate and valerate molar proportions, lower (P < 0.01) acetate and acetate:propionate ratio than steers fed LQ diets. In vitro methane and total gas production were not affected (P ≥ 0.50) by dietary treatments. Fibrolytic enzymes positively affected digestion of multiple roughage sources commonly fed to cattle and might have additional benefit when used on unprocessed sorghum grain. Fibrolytic enzymes in beef cattle growing diets stimulated intake and generated positive impacts on ruminal fermentation.

Two treatments were evaluated in heifers to determine the effects of a yeast supplement on immune and metabolic responses to a combined (tandem viral-bacterial) respiratory disease challenge. Thirty-two beef heifers (325 ± 20.1 kg BW) were selected from a larger population previously assigned to one of two treatments: Control (CON), receiving no yeast supplement in the diet, or yeast (YST), CON diet plus a combination live yeast (2.5 g·heifer-1·d-1) and yeast cell wall (2.5 g·heifer-1·d-1) supplement (Phileo Lesaffre Animal Care, Milwaukee, WI). Heifers were maintained on treatments for 31 d prior to the challenge. On day -3 all heifers were fitted with an indwelling vaginal temperature recording device, received an intranasal challenge with 2 × 108 plaque-forming units of bovine herpesvirus-1 (BHV-1), and placed in outdoor pens. On day 0, all heifers were fitted with an indwelling jugular catheter, challenged intratracheally with an average dose of 3.0 × 107 colony-forming units (cfu) of Mannheimia haemolytica in 100 mL media, and were transferred into individual stanchions in an enclosed, environmentally controlled barn. Whole blood samples were collected at -72 h and at 0, 2, 4, 6, 8, 12, 24, 36, 48, 60, and 72 h (hematology) and at 0, 1, 2, 3, 4, 5, 6, 7, 8, 12, 24, 36, 48, 60, and 72 h (serum isolation) relative to M. haemolytica challenge (0 h). Data were analyzed using the MIXED procedure of SAS specific for repeated measures with fixed effects of treatment, time, and the treatment × time interaction. Vaginal temperature and cortisol concentrations were similar between treatments (P ≥ 0.39). Although total leukocyte count following BHV-1 challenge was similar between treatments (P = 0.21), there was a tendency (P = 0.07) for CON heifers to have greater neutrophil counts than YST heifers. Serum haptoglobin concentration was similar between treatments (P = 0.13). Heifers in the YST treatment had similar serum glucose concentrations (P = 0.25) and decreased serum concentrations of urea nitrogen compared to CON (P = 0.03). Dietary treatment did not affect serum nonesterified fatty acid concentrations (P = 0.37). Nasal lesion score severity (0-4) tended (P = 0.07) to be decreased in YST compared to CON (2.5 vs. 3.19 ± 0.26), while water intake tended to be increased in YST (P = 0.06). Feeding a yeast supplement had little effect on the acute-phase response but improved metabolic outcomes in heifers during a respiratory disease challenge.

OBJECTIVE To investigate the effects of meloxicam administration before long-distance transport on inflammatory mediators and leukocyte function of cattle at feedlot arrival. ANIMALS 60 healthy yearling beef steers. PROCEDURES Single-source steers were assigned to a transported (n = 40) or nontransported (20) group. Then, half of the steers within each group were assigned to receive meloxicam (1 mg/kg, PO) or a lactose placebo (1 bolus/steer, PO). All steers were transported approximately 1,300 km overnight to a feedlot; however, the nontransported group was moved before treatment (meloxicam or placebo) administration and allowed a 17-day acclimation period, whereas the transported group was moved immediately after treatment administration on day -1. Blood samples for measurement of inflammatory mediators and leukocyte function were collected from all steers on days -1, 0, and 3. RESULTS For steers that received meloxicam, mean plasma meloxicam concentration for the transported group was significantly greater than that for the nontransported group on day 0. For steers that received the placebo, mean haptoglobin-matrix metalloproteinase-9 complex for the transported group was significantly greater than that for the nontransported group on day 0. Mean haptoglobin concentration, neutrophil L-selectin intensity, and polymorphonuclear leukocyte count for the transported group were significantly greater than those for the nontransported group. Mean substance P concentration for nontransported steers that received meloxicam was significantly lower than that for the other 3 treatment groups. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated meloxicam administration to healthy steers immediately before long-distance transport did not significantly mitigate the effects of transport-induced stress on leukocyte function or inflammatory markers.
